Assuming the adaptation does what adaptations usually do and diverges from the source at various points (like Preacher itself does) rather than following the original precisely, they'll definitely be classified as separate sources.

(I honestly can't think of any comic adaptations that do mirror the source closely enough to count as one and the same, but I figured I'd add that qualifier in there just to be safe.)
